Accurate or precise? When does my pipette need to be calibrated?

Once or regularly? How often does my pipette need to be calibrated?

Calibration is a gravimetric check of pipettes. Regular calibration is used to determine and correct the difference between the dispensed volume and the set volume. Due to frequent use and pipetting of different liquids, the pipette may not perform accurately over time. This depends on several factors, including whether it is an air cushion pipette, a direct displacement pipette or a stepper. According to the manufacturer's recommendation, a pipette check should take place between 3 and 12 months, depending on the frequency of pipetting operations and the type of liquids pipetted.

The main reasons for not doing your pipette calibration yourself are high costs for equipment, personnel and special environmental conditions. Here we have compiled the most important points for you in detail!

#1 Air-conditioned measuring room

The design and working principle of air cushion pipettes require defined environmental conditions:


  • Temperature
  • Relative humidity
  • Air pressure


These parameters have a direct influence on the conditions in the air cushion and cause large errors if they are not exactly set or known. The room conditions may only deviate minimally from the setpoint, otherwise no calibration is possible.

#2 Measurement sequence is predefined and must be followed exactly

An example of this is the following sequence:


  • Pipette tip must be conditioned with the test liquid (distilled water), aspirate and dispense 5 times
  • Aspirate set volume and wait briefly
  • Dispense volume into liquid and keep thumb on aspirating button while pulling out
  • Weigh
  • Hold pipette vertically while aspirating liquid and withdrawing tip from test liquid. Maintain a 30-45° angle when dispensing liquid

#3 Accurate measurement procedures

Extremely accurate measuring instruments are required to ensure the necessary accuracy classes of a successful pipette calibration. The following requirement profiles must be met:
