Western blot membranes must bind proteins and antibodies optimally so that even low protein concentrations can be detected effectively. However, due to the binding properties of the membrane surface, insufficient blocking of the free binding sites after protein transfer leads to non-specific background signals and erroneous results. For clean blots and reproducible, reliable results, suitable blocking is therefore mandatory. Depending on the goal of the application (routine; quality control; publication; diagnostics) and the type of sample, different types of blockers are available. For many standard applications, the inexpensive milk powder or albumin (BSA) is the blocker of choice. However, for high background and non-specific bands, a switch to more potent alternatives is recommended.
